Can the wheels be replaced, meaning do they come off?

Yes they could. I got better heavy duty wheels for mine. They work great.

No they do not lock. It’s a very nice sturdy stool.

It's more of a leather like material.... but looks great.

Do the wheels plug into plastic or metal holes?

Plastic. I upgraded to metal inserts and bigger wheels.
