Yes you can use a brush. I would test on both to see which way works best for you.

Q: Can this ink safely be used in fountain pens?

I have never used it in fountain pens, only in Rapidograph pens.

Q: Is this a pen, or has a removable top ? I am looking for something I can mix with pigment powder on a canvas so I need to pour it out.

No this is not a pen. It is ink that you fill a rapidograph pen with and I think it would be quite messy to add it to pigment powder but it can be poured out. I am curious how you would mix it with pigment powder to apply it to a canvas?
