Q: Why is there only one strain in these ?

Not really a question other consumers can answer. Their liquid probiotic has many strands and tastes like water. You get more bang for your buck with the liquid version. My kids 3, 5, 7 all like it.

Q: Is there a sugar-free version?

Take a look at the liquid version they offer. In all of my searching, Mary Ruth offers the best sugar ratios on the market (if you want ingredients you can trust)

Q: Is it strange that these have been sold out on their website but always available on Amazon?

I'm guessing they may have allocated a certain amount for Amazon to keep in stock. If so, it wouldn't be strange.
